I agree, travel at night. But if that is not possible (I know how hard it is to drive at night) I would think this should work.
As for the 3 year old, I would bring a portable DVD player, some car games, paper & crayons, something to play on (say a tray of some sort).
As for the 5 week old (unless he is bothered by traveling), he may well sleep during most of the trip (except for feedings & changes). I know our 2 month old did when we travelled 6 hrs with her.
At any rate plan several stops (don't make it scheduled stop, 3 year olds & 5 week olds don't work on anyones schedules). It may also help to travel part way during the day (say 4 hrs or so) and the balance into the night. That way the driving parent won't have so hard of time travelling all night.

Most pediatricians recommend liquid Benedryl 30 minutes before take off and then a bottle, breast or pacifier as the plane is taking off and landing. The pressure does bad things to the baby's ears unless she's forced to suck. As for entertaining her during the flight. Good luck. It's a long one.
